Instrument Processing
Once the gStack is loaded, all dilutions, mixing, incubations and reads are completely unattended. The first operation is the removal of six of the twelve gCards from the gStack and placement of the gCards on the spindle.

Each card contains a network of microfluidic channels, and the liquid is driven through these channels using the centrifugal force generated by the rotating spindle. While the spindle rotates, an optical system performs two functions:

• It actuates a series of Virtual Laser Valves (VLVs), thereby determining dynamically and accurately the volume of liquid moving from one point to another.
• It provides a kinetic readout of the fluorescence from each data point which has been set up.
